How would you define, Rational Drug Design?

discovering the specific shape and charge of a substance you wish to target (IE: enzyme active site)
producing a drug complementary in shape and charge the substance (IE: enzyme active site)

Also, what is the best way to go about doing one of the 'design an experiment' questions.
Some help would be appreciated :)

^ Yeah they're a pretty big pain in the rear if you ask me...Knowing the tips of what to write would be handy! Hopefully someone replies!

experimental design?
how i got about it, i do it in dot points
- 2 large groups (100+) of genetically similar mice (example), group A is the control, group B is the experimental group
- talk about controlling variables, then say EG: food and water (apparently if you don't say EG or IE or example some examiners thing that you think there the only variables.) then talk about giving both groups the infection (if its an infection) then talk about giving the group B the drug (if its a drug of course). if it is something that after you give the groups and it takes a while, you need to state 2 weeks for example
- talk about observing and recording the results and repeating the experiment for increase accuracy in the results
- then state if you got such and such results how that would support/negate the hypothesis
