I'm quite sure that this is possible, because if you look at the irregular offer form, the selection officer has to tick a box stating whether the offer was made in round 2, or after round 2.
However, second round offers are made on a "first come, first served" basis, so you are more likely to get an irregular offer if you apply for one before 2nd rounds come out.
