If I received by first choice for the first round (which was preference no. 1), would it still be possible to change my preferences and get an offer for a course that was initially lower than my first round?

Yeah, I'm pretty sure you can move your preference priorities around tomorrow. Accept your course and then shuffle them around and pray for a 2nd round offer.

What are you interested in changing into, out of curiosity?
2016–2018: Bachelor of Biomedicine (Neuroscience), The University of Melbourne
2019–2022: Doctor of Medicine, The University of Melbourne

Even with shuffling of preferences, does this still mean that I retain my initially accepted offer regardless of whether I receive my other. In other words, would I be risking losing that first offer?

Edit: Commerce at Melbourne btw, looking to move into post-grad Engineering. I'm looking at the change namely for the location.

I'm in a similar situation, and I was just hoping somebody could clarify for me. Basically I've realised that I should have chosen my second preference over my first, but I was just told by my sister that the chances of receiving a second offer is low. Does anyone know whether this is true or not?? My second pref is Monash Arts.
ALSO, does anyone have any experience/knowledge on how to accept a second preference over a first preference???

Yes. Put the course you want the most first.
Graduated with Bachelor of Laws (Honours) / Bachelor of Arts from Monash University in June 2020.
