what did you guys write for the YAC and why the centromere?

Ahahahaha
ahahahaha
I was like UMMMMMMMM.
I put something about it being there to hold the allelic DNA and the whatever the other thing was in place because that chromosome was a free and isolated one, whereas the plasmid didn't need one because it's circular and therefore all DNA will be held in place without a centromere.

Yeah.  I had no idea.

If I understand this question correctly, what others have said regarding spindle fibres is correct. In prometaphase (midway between prophase and metaphase), the spindle fibres from the centromeres attach to proteins located on either side of the centromere, and in anaphase after the centromere is cleaved, these fibres then separate the two sister chromatids. This isn't required in bacteria because their DNA reproduces by binary fission instead of mitosis.

I agree with the spindle fibers interpretation also.  I seriously had no clue, and I swear we were never taught that the spindle fibers from the centromeres are required for cell division in my class.  I was just like 'think about what's different between plasmids and linear DNA', but I guess what I really should have thought about is what's different about eukaryotic cell division and bacterial cell division.  Oh well. It was one mark or something, I'm p. sure.

shiny, do you think my answer for the YAC question is right i wrote: "The Yac has a centromere as it is required in cell division".

Hard to tell without knowing what the stem for that question was, and the mark allocation for it too. That'd probably get at least 1 mark though I guess.

for that "what is a probe?" question, what kind of probe were they talking about?
I wrote: A strand of RNA complementary to the strand of DNA being sought :S
Is that even remotely correct?

Single-stranded DNA or RNA with a base sequence complementary to that of a target strand of DNA, and that carries a fluorescent or radioactive label for its detection.

I wrote that it'd be black, cos it'd be the same as the mother's. But really, I have no idea.

I said breast cancer can be genetic, so the daughter strand could be red. But I was pretty confused about the last question overall.

i thought it'd be red because the daughter inherits a mutant allele from the mother :S no ideaaaa

i said it could have been red or green depending on whether or not the mutaiton afected germline cells. if it did it would be red, if not it would be green and healthy!
