EDIT: Why am I so dumb? Haha of course they do but I forgot to realize that post-transcriptional modification of the primary transcript or pre-mRNA would simply remove this amino acid if it is not vital for the polypeptide's primary structure

For the latter part of your question, recall that genome varies for different species (e.g. human genome, wallaby genome etc.) hence your first part is correct.
For the first part of your question have a read of your text-book for exact definition and it is not the same as introns because introns are transcribed whereas spacer DNA is not transcribed.
